Pineapple Express was boring on the second viewing, the nail in the coffin for a comedy.

Agreed. It's not just perhaps THE single worst comedy I've seen in recent years, but most likely one of the worst movies I've seen, period.

Half the comedies mentioned in this thread so far involve jokes about sex and genitals.

I think the Hangover is going to endure. And from Apatow's films, Anchorman will probably be most fondly remembered, followed by 40 year old virgin.

OKAY. Let's be aware that it's already 2012. Some of the films listed on this thread are old enough that we can actually judge if they have endured or not. Let's go by year.

2000
1. Meet the Parents - Not as popular as it was 12 years ago. But it is one of the more fondly remembered Ben Stiller films.
2. Scary Movie - The sequels ruined the franchise, but everyone still loves the original. It is one of the more popular spoof movies of the 21st century.
3. Miss Congeniality - Girls still love this movie. Obviously the CS! female population is practically non-existant, so I have to be the one to bring it up. This film - definitely a classic - at least for females.
4. ME, MYSELF, & IRENE - It's a classic. PERIOD.
5. Bring It On - Raise your hand if you're a pervert. Elizha Dushku. Car Wash scene. CLASSIC.

2001
1. Shrek - CLASSIC
2. Monster's Inc - CLASSIC
3. Legally Blonde - It got a Broadway adaptation. So... what do you think?
4. Bridget Jones' Diary - Alien. Deexan. Where you at?
5. Zoolander - CULT Hit.

2002
1. My Big Fat Greek Wedding - Indie Classic. For sure.
2. Maid in Manhattan - Got adapted into a successful spanish soap opera.
3. Barbershop - Any Black people here? Besides me, of course.
4. Super Troopers - CULT Hit.
5. Secretary - CULT Hit.

2003
1. Finding Nemo - CLASSIC.
2. Elf - Holiday Classic.
3. School of Rock - CLASSIC.
4. Old School - CULT Hit.
5. Bad Santa - Holiday Classic.
OVERALL: Darn good year. And I didn't even scrap the surface.

2004
1. Shrek 2 - CLASSIC.
2. Means Girls - CLASSIC.
3. Anchorman - CLASSIC.
4. Sideways - Indie Classic.
5. Napoleon Dynamite - Not as successful as in `04, but it has enough of a following to get a cartoon version all these years later.
OVERALL: Another good year. And again, didn't even scratch the surface.

2005
1. 40 Year Old Virgin - CLASSIC.
2. Wedding Crashers - Hard to say. It was definitely more popular in `05, but still has a following of sorts.
3. Charlie and the Chocolate Factory - There's definitely a generational gap with this film. But I can assure you that today's teens, who were kids in 2005, LOVE this movie. And the kids of TODAY also love it. Fans of the Gene Wilder version don't want to acknowledge it, but it's a reality that has to be faced.
4. Jarhead - It's a dark/satirical comedy. I think it's still relevant.
5. Kung Fu Hustle - Definitely big in the Asian market.

2006
1. Borat - CLASSIC
2. Talladega Nights - CLASSIC
3. The Devil Wears Prada - Big with females.
4. Little Miss Sunshine - Indie Hit
5. Clerks 2 - Kevin Smith has a strange following.

I'll stop here because everything else is too recent to judge.

You generally seem to have very, very, very, very low standards for what constitutes a "classic."

UM,,yeah

Classics have to be at least 20 years old just to begin with.

Plus Ian said "straight-up" comedies, which many of those movies are not.

I would like to throw 'I Love You, Man' a bone, it's probably the best comedy I've seen it the last 5 years.

Yeah, but we're speculating which ones will be, so lets put that "not old enough to be classic" argument to the side for now.
